Abstract:
Vibrational sum-frequency generation (VSFG) spectroscopy is a nonlinear optical technique that has emerged as a valuable tool for studying the structure and dynamics of molecules at the interfaces. Through this technique we can get a detailed information about the vibrational modes of molecules, specifically at the interface, which is difficult to attain via Raman and IR spectroscopy due to strong scattering and absorption from the bulk of the material.
In this talk, first I will explain the fundamental concept of VSFG process like how the sum frequency is generated from the visible and broadband IR light beams. Next, I will explain the detailed optical setup to generate polarisation sensitive, high intensity VSFG signal at the interface. Then I will explain how the VSFG technique can be utilized to understand the misfolding mechanism of proteins. I will give a specific example of the misfolding process of human Islet Amyloid Polypeptide (hIAPP) which is responsible for type 2 diabetes. Finally, I will conclude my talk by giving a brief introduction of time resolved VSFG technique that is helpful to understand the dynamics of vibrational energy transfer (VET) processes of proteins at the cell membrane.
